From what I seen so far normally vacuum lone normally is located on the edge (opposite side to resin feed).Is that the best way to do it?
Is the resin should travel on the shortest route?

Yes, choosing the shortest route is usually the best option Depending upon the mould shape, you might also need to give some thought to the distribution of the resin to try and get the part evenly.....
