From what I seen so far normally vacuum lone normally is located on the edge (opposite side to resin feed). Is that the best way to do it?
Is the resin should travel on the shortest route?

Yes, choosing the shortest route is usually the best option

Depending upon the mould shape, you might also need to give some thought to the distribution of the resin to try and get the part evenly wetted-out prior to the resin reaching the outlet port

As an example, on wide moulds with a short distance between the inlet and outlet, you would look to use resin spiral to quickly distribute the resin feed to the farthest edges.
As the resin would likely reach the outlet before reaching all of the outer sections, some consideration needs to be given to slowing things down a little around the centre to allow things to infuse as evenly as possible. This is where a bit of planning in the placement and cutting of the flow media can help (using resin breaks etc.). Doubling-up of the flow media in certain areas can also help to speed things up. It's on bigger moulds approaching 1m wide that this might be helpful, especially
