I've miss calculated the amount of resin needed for my mould and haven't achieved 4 layers, is there still an option of applying more within a time frame? or is it best to wait and key in and add more??

How many layers did you get down? You can apply more layers within a 24 hour time frame, though I would scuff up the surface with 60 grit paper and clean well with acetone regardless. If longer than 24 hours, make sure you scuff up and clean.
The problem with tooling resin is it actually needs to 'self-bake' to achieve its full properties, and it needs a minimum of 3 layers of 450g csm to do that. If you didn't get that down, then are you able to apply more layers and do an elevated temp cure? If you can get it up to 60 degrees minimum then you may get away with it.

I only got 2 layers on as there wouldn't of been enough resin to complete the third layer, so stopped at 2 layers. That's what I worry about now is the self bake. How long for a 60° bake? I should be able to do this

You do not need much time at 60 deg .. The unimoulds I have made were probably at 60 degrees for less than an hour ..
