I did not read any scientific work about it, but I think the main reason two sides are used is that prepregs are often cut on plotter tables, wich need both sides protected to stay relatively clean. And the precuts can be piled easily and don't pick up dirt during the process. Some prepreg impregnation processes even utilise two separation films, as the resin is worked in the fabric by pressing the prepreg between two rollers several times.
