OK, that should be OK. If the filler is polyester filler (whcih most of them are) you will need to ensure that the polyester filler is completely cured and is no longer smelling of styrene. If it is, you should wait a while for it to fully cure off and stabilise a bit otherwise you run the risk of the filler upsetting the release agent (whether it's wax or something else).
In terms of your paint finish, it's fine to do this but please make sure you use a two-pack / 2k paint (polyurethane or acryllic) NOT a rattle-can spray paint. We have endless horror stories from customers who have used rattle-can spray paint on their patterns and then have lots of problems releasing moulds.
