If you are perfectly happy with the shape, then you can go over with high gloss and it should seal the surface no problem as long as the spots of MDF aren't too porous. Worst case you might have to do a second coat.
In future, what helps is to pigment each layer of Pattern-Coat Primer so that when you sand through to a layer below, you can clearly see that you are doing it and adjust your sanding technique accordingly to avoid breaking through to the substrate.
