Kitchen wrap or release film would be your best bet... Thats a good idea and probably how I would do it.

Simply, how to make a mould from a piece covered by leather, without causing damages to it (and to be able to restore it) ?
Any sort of protection to lay up first which would be harmless to the leather, before applying ease-lease release agent, wax, and gel coat ?

I have made polyester molds over wet clay and over styrofoam. There are 4 ways that you can do this, that has worked for me.
1. I cover the piece with saran wrap. (kitchen wrap the cling kind) carefully gelcoat and lay up your reinforcement when tacky.
2. Tin foil over the piece. I will often spray a little tackifier (3M 77) it get it to stick. Make your mold.
3. Packing tape over the whole piece and then wax. Now make your mold.(it may tear your leather when you release the tape so the above might work better.
4. Use silicone rubber. it will release cleanly and have every detail.
